Tasks to develop
1- Identification of adipocyte secretion pattern – different depots and different conditions
Objectives:
• Do an extensive analasys of the adipocyte transcription and secretion profile during obesity etiopathogenesis.
• Detection of the differences in adipocyte function and secretion due to depot localization, sex and age.
Implementation:
Month 1 – Month 42
Deliverables:
• Identification of the adipocyte transcriptome and secretion.
• Characterization of differences in adipocyte function and secretion due to depot localization, gender and age.
• Descovery of new adipokines.
2- Molecular mechanisms of beta cell adaptation to obesity
Objectives:
Study the changes in the islet gene expression during the progression towards obesity
Implementation:
Month 1 – Month 36
Deliverables:
• Characterization of the islet adaptation to an hypercaloric diet and insulin resistance.
• Identify the molecular mechanisms of response to a reduction of insulin resistence in an obese animal, role of insulin signalling pathway.
3- Identification of adipokines involved in beta cell replication and apoptosis
Objectives:
• Identify the factors secreted by adipose tissue which are able to control beta cell replication and apoptosis
• Characterize the adipokine-mediate crosstalk with the beta cell.
Implementation:
Month 7 – Month 48
Deliverables:
• Identification of the adipokines that control the beta cell mass homeostasis
• Characterization of the molecular mechanisms of beta cell function (replication, apoptosis)
• Integration of collected data for different beta cell phenotypes and states.
• Identify, a limited and manageable subset of those genes/proteins predicted to offer the greatest promise as potential therapeutic targets.

Deliverables:
Report on a list of genes/proteins to be used as potential therapeutic targets for obesity.
